Diversified application of microfluidic chip in cell research
A creature is everything that has metabolism. Narrowly defined organisms are objects that are traditionally independent and capable of autonomous survival, including animals, plants, and microorganisms. The organism has the characteristics of inheritance and variation, can grow, develop and multiply, can adapt to certain environments and change the environment, and can respond to external stimuli. Cells are the basic unit of structure and function of most organisms. The microfluidic chip technology developed in the 1990s has broad potential in cell research and has attracted great attention. At present, the application of microfluidic chips in the field of cell biology mainly focuses on cell response and cell behavior in the microenvironment, and its research scope covers many biological species. Below we describe the application of microfluidic chips in the field of cell biology from three representative aspects of animal cells, plant cells and microbial cells. First, animal cell chip The application of microfluidic chip technology in animal cells is very extensive, especially in mammalian cells. In addition to basic cell culture, the researchers also carried out a large number of microfluidic chip-based normal cell and tumor cell manipulation, analysis and practical application research, including cell classification, cell fusion, single cell capture and gene analysis, cells and micro Environmental interactions, high-throughput drug screening, etc. A large number of cell analysis applications based on microfluidic chips are emerging, greatly improving the efficiency of cell analysis. For example, Sugiura et al. prepared a parallel array microcavity cell chip that can simultaneously study the anticancer effects of seven antitumor drugs on human cervical cancer Hela cells. Second, plant cell chip The variety of plant kingdoms varies widely. Similar to animals, higher plant individuals are also composed of a large number of cells. The division of labor between the various cells and the differentiation of morphological structure, interdependence and cooperation, together to maintain the normal life activities of the entire organism. At present, there are few reports on microfluidic chip technology in plant cells, but plant cells have many similarities with animal cells. For example, plant protoplasts (ie, plant cells that remove cell walls) have many similarities with mammalian cells. Large molecular substances such as organelles, microsomes, viruses, and DNA can be taken, and are often used for plant breeding research in various membrane biology and cell-based fusion technologies. In 2010, Wuheng Institute firstly used PEU chemical fusion method to achieve microfluidic chip fusion of protoplasts of tobacco seedlings and protoplasts of tobacco mesophyll. Third, microbial chips Microorganisms include a large group of organisms such as bacteria and fungi, which are closely related to human life. Microorganisms cover a wide range of beneficial and harmful species, including human health, food manufacturing and safety, medicine and health, and environmental protection. At present, the application of microfluidic chips in microbiology mainly includes two types: one is the study of microbial characteristics, behavior and individual component analysis (see the figure below), such as Kim et al. using a concentration gradient chip. Experimental study on drug sensitivity of various antibiotics based on bacterial biofilms. The other is the analysis of microbial properties, behavioral and individual components, such as the Whitesides team's research on the behavioral behavior of E. coli using microchannels.
